There a 3 major players in 3rd party products for Des2k, the two you mentioned and Guidelines/2000. I researched all 3 for my company and we decided on DesignAssist, and its been a great decision. We developed an much more attractive and flexible system using the tool.
Headstart is similar to DesignAssist in that it is a collection of templates and packages, however you must purchase a certain amount of Oracle consulting to get the product. Guidelines/2000 is very much geared toward collecting information in the analysis phase of development, so if your already into design...forget about it. It also uses repository extensions to implement its features, and most people will recommend staying away from that side of Des2k.
I believe the price on DesignAssist recently dropped, so it would be my recommendation to go for it. You'll definitely save time.
