I currently develop applications in both PowerBuilder EE 5.0 and VB Professional 4.0. Based on my experience I believe both development environments to be highly useful. The major difference being that native database access is MUCH easier with PB. If the application is a "Reporting" based app I will choose PB. If limited SQL/ODBC programming is required I will use VB. If I had to choose only one environment I would choose PB. Although I'm glad I don't have to choose.
