First the correct spelling is SQLNET.EXPIRE_TIME and since the parameter goes in the sqlnet.ora file I believe that it will take immediate effect for every session started after the file is updated without a listener or database shutdown being necessary. However, it will affect only new sessions started after the file change and will not affect already connected sessions.
