I follow a very strict and simple rule DDL after hours and DML during business hours. Sure, you can change RI, but as I troubleshoot errors it frustrates me to see changes imposed mid-business day. You can also through new entire sub-systems into a database while the user is on, but what do you do if you mess up on sizing and choke out the user temp spaces for example. I have a DBA that chooses to not work after hours and "sneaks things in". I always say pay me now or pay me later because it will bite you.

The answers above are all correct, you can't safely alter objects so their knee jerk reaction is to get out of the way impending disaster and deal with file sizing/optimization first opertunity off-line hours.
