Hi,
RAC has no additional role to play in migrating from file system to ASM. ASM is mandatory for RAC but not the otherway around.

what is the oracle db version and how big is your database?. approach that you will choose will depend on the size and the down time you will get. you could do a simple RMAN backup and restore to migrate to ASM.

RMAN backup would be a byte to byte copy of the original database and would not help you in defragmenting.
