I m not able to run tkprof from >sql command promt
is it nessassory to run any script file before using tkprof?

What do you mean by "I'm not able to run TKPROF"? You will have needed to have generated a trace file first and this filename would then be passed as a parameter to the TKPROF utility.

but its showing error unknown command beginning from tkprof

tkprof is a command line utility NOT SQL*Plus.
